In a study involving mice, deactivating the PV-1 gene prevented the normal metabolic shift and the liver continued to prioritize carbohydrate burning while redirecting fatty acids to muscles rather than being processed by the liver.

The GLVAP disruption maintained high metabolic activity without the mice’s bodies entering the typical "starvation mode" associated with calorie restriction. The study’s surprising breakthrough in metabolic efficiency suggests renewed potential for finding innovative solutions to frustrating weight-loss plateaus that are commonly faced by many obese or overweight individuals who need to lose substantial amounts of weight.

Collaborators from Japan, the USA, and Finland. Danish scientists suggest that targeting the PV-1 gene could be key to overcoming common barriers by offering new hope for those with significant weight-loss goals.

How Cells Trigger Metabolic Changes 

The PLVAP gene is expressed in hepatic stellate cells, which are now recognized as key players in metabolic regulation. By disabling the gene in mice, researchers found that the liver could no longer recognize the fasting state. Instead, fat was redirected from the liver to muscles, allowing the liver to continue improving insulin sensitivity without adverse effects. This unique discovery suggests PV-1 is vital in determining whether the body burns sugar or fat during fasting.
